5 Performing a Volume Transfer
To transfer the eluates of a purification run into the wells of another plate suitable for e.g., a PCR/RT-PCR
run, you can use the volume transfer order type.
The volume transfer run comprises the following steps:
1. Defining the parameters in the software, see section Defining the Parameters in the Software.
2. Loading the instrument, see section Loading the Stage.
In this example the eluates reside in the MagNA Pure 96 Output Plate (defined as source plate in position
4 of the purification rack) and are transferred to a LightCycler
®
480 Multiwell Plate 96 (defined as target
plate in position 3 of the purification rack).
Automated liquid handling systems like the MagNA Pure 96 System require more volume than the ef-
fective volume to ensure exact pipetting results.
When calculating the volumes for the volume transfer function, the MagNA Pure 96 Software considers
the dead volume. Consequently there will be some residual liquid left in the respective wells or tubes of
a plate.
Only the number of tips corresponding to the number of samples are used for the volume transfer pro-
cedure.
5.1 Defining the Parameters in the Software
Before starting a volume transfer run, the operator must provide the software with the necessary informa-
tion on protocol and sample parameters. The most important and obligatory information is the source
plate type and the transfer volume.
You have several possibilities for selecting the plate:
Select the source plate directly via the barcode reader of the reagent head. To use this function the
source plate must be placed on the stage.
Scan the barcode of the source plate with the handheld barcode scanner.
Enter the barcode of the source plate manually.
The following steps can be performed by users with the standard user or local administrator role.
