To resolve unstable fluidics (air or clog in system) in Luminex Measuring Instruments, first, ensure a sufficient sample volume is used; 50 ?l is recommended. To clear any air bubbles, run the purge bubbles script. If detergents or foaming agents are causing bubbles, remove them and re-suspend the sample in dPBS, then run the purge bubbles script. For air bubbles in fluid lines, run the sterilize script, followed by the startup script, then load calibration beads to verify the system runs normally. If there's a clog in the fluid lines, filter the sample with a 70 ?m nylon cell strainer, then run the sterilize and startup scripts, and verify with calibration beads.
