      To resolve unstable fluidics (air or clog in system) in Luminex Measuring Instruments, first, ensure a sufficient sample volume is used; 50 ?l is recommended. To clear any air bubbles, run the purge bubbles script. If detergents or foaming agents are causing bubbles, remove them and re-suspend the sample in dPBS, then run the purge bubbles script. For air bubbles in fluid lines, run the sterilize script, followed by the startup script, then load calibration beads to verify the system runs normally. If there's a clog in the fluid lines, filter the sample with a 70 ?m nylon cell strainer, then run the sterilize and startup scripts, and verify with calibration beads. If the sample is too concentrated, dilute it to 1x10^7 cells/mL, strain it through a 70 ?m nylon mesh, and run the purge bubble...

      If INSPIRE™ appears to freeze on Luminex Measuring Instruments, first, check if the camera is running by clicking Run/Setup. If the camera is already running, try clicking Stop and then Run/Setup again. If imaging is paused, click Resume. If there are no objects in the current image view mode, select the all population in the cell view area. Lastly, if a script is running, wait until it completes, or click Abort Script to prematurely stop the operation.

The sheath syringe should contain 2-4 mL of air to buffer the movement of the pump’s microstepper motor. If too little air is present, run the “start-up” script. Another possible cause is leaking fluid lines. In this case, look for leaking sheath with the system powered down and verify that the fluid lines mount snugly into position. If the issue persists, contact Luminex Technical Support.

      If INSPIRE™ fails to launch on Luminex Measuring Instruments, and the splash screen is not responding, press Ctrl-Alt-Delete on the keyboard, open the task manager, select INSPIRE™ for FlowSight® Software, and press end task. Wait 60 seconds and try restarting INSPIRE™. If there is a loss of communication between the computers and instrument, shut down the computer and power off the instrument. Verify all computers are off, then power on the instrument and then the computer. Wait 5 minutes and try launching INSPIRE™.

If calibration particles are not running properly, and the beads are diluted or clumped, try running a fresh tube of beads. Finally, if there is a calibration and/or test failure, re-run the test by clicking in the box next to the test and pressing the start button in the popup window. If the test fails three times in a row, contact Luminex Technical Support.
