Step 3. Prepare the performance evaluation samples
In this step, you dilute the supplied performance evaluation sample
(reserpine) to the concentrations needed for the Triple Quadrupole LC/MS
system checkout.
Use the diluted samples within a day of dilution. Refrigerate the intermediate
(first) dilution in the supplied bottle.
• Always rinse the graduated pipettes and volumetric flasks thoroughly with
deionized water before and between each use.
• Use polypropylene labware for preparing performance evaluation samples,
since glass vessels introduce unacceptable levels of sodium.
• Always rinse the autosampler vials and caps with the solvent mix used for
sample dilution before filling them with the performance verification
samples. This minimizes any background that can be contributed by the
vials and caps. The vials may be run uncapped if the septa are found to be a
source of background contamination.
Negative Mode (all models except 6410)
Required Parts • 1 mL graduated pipette (p/n 9301-1423)
• 2 × 100 mL volumetric flasks (p/n 9301-1344)
• ESI Negative Ion Performance Standard (p/n 5190-0591)
• Plastic bottle for storing first dilution (p/n 9301-1433)
1 Prepare 2 liters of a 70:30 mixture of water:methanol.
2 Prepare the first dilution (50 pg/μL) chloramphenicol:
a Use a clean graduated pipette to transfer 1.0 mL of the 5 ng/μL
chloramphenicol (p/n 5190-0591) standard to a pre-rinsed 100 mL
volumetric flask.
b Dilute to the 100 mL mark with the 70:30 water:methanol solution that
was prepared in step 1. Invert the flask at least 10 times to ensure proper
mixing of the checkout sample.
c Use 5 to 10 mL of this solution to rinse one of the plastic storage bottles
and fill it with the remaining solution.
d Label this bottle “50 pg/μL chloramphenicol” along with the preparation
date.
