A.4. Skin sample for MOHS with glass disc technique
The difficult part in skin sample freezing is to put it with all its margins in the same level and so in one slide.
Milestone studied a method that, through the use of glass, allows to fix all the margins to the flat glass, in
order to have them at the same level in the frozen block.
1) Relax and ink the sample, following the standard lab procedure.
2) Put the glass disc inside the dedicated spot of the hand protection tool (available only for 22 and
32mm discs) or hold the 48mm disc directly with the fingers.
3) Put the sample on the glass disc with the epidermis up and in the needed position. Push down and
adhere sample margins to the glass.
4) If necessary, press down one site of the margin with the finger and freeze it, cryospraying the
corresponding position from the other side of the glass. Repeat for all the margins.
5) Put the glass disc with the sample inside the PrestoCHILL mould and fill the mould with MCC.
6) Put chuck and heat extractor and freeze at -40°C for the suggested time for the used mould (see
appendix B).
7) Detach the glass disc from the block, heating it with the palm for few seconds.
8) Trim and cut in the cryostat.
