C. Appendix – Use the extractor for chucks
At the end of freezing the block could stick to the mould, as a result of one of the following factors:
• Frost inside the mould before its use. In this case ice forms between the block and the mould that
acts as glue between the two parts.
Cleaning the mould is very important to prevent the block from sticking to the mould.
• Use of warm embedding medium (temperature above 25°C). In this case ice may form in the
embedding medium layer in contact with the mould.
• Using moulds deeper than 5mm: when preparing specimens in 7 and 10mm deep moulds it is more
difficult to extract the block because the contact surface between block and mould is larger.
• Using embedding medium other than Milestone Cryoembedding Compound.
Milestone recommends using embedding medium Milestone Cryoembedding Compound
(MCC - Code 51420).
For optimal results, Milestone recommends using 5mm deep moulds.
C.1. How to use the extractor for chucks
• Loosen, by rotating anti-clockwise, the jaw stop, so that the jaws widen more than the diameter of
the chuck handle
• Insert the jaws under the chuck handle
• Close and secure the jaws by rotating the stop clockwise
