To create the order
Open the load flap and pull the purification rack all the way out of the stage.
Handle the purification rack carefully because of its imbalance. Do not place it on the load flap, to
prevent it from falling off.
Place the MagNA Pure 96 Output Plate with the eluates purified by the same instrument before in posi-
tion 4 of the purification rack.
Insert the purification rack back into the stage and close the load flap.
Insert the rack carefully so as not to spill the eluates onto the stage.
Wait until the barcode scan has finished.
Open the Workplace tab in the MagNA Pure 96 Software.
On the Workplace tab select the Orders sub-tab. The Orders sub-tab opens.
Select Volume Transfer from the order type list. The Parent group is displayed.
Click the Use Onboard Plate button to read the barcode of the source plate. If reading is successful the
software displays the Test Parameter area and
The barcode in the Plate ID field
The plate type in the Source Plate field
The volume present in the Volume Source field
The names of the eluates in the sample table
In the Test Parameter area, select a PCR Setup protocol from the Protocol list:
Select the PCR Setup with Mix protocol if transferring eluates that have been stored at -15 to
-25 °C or -60 to -80 °C.
Select the PCR Setup without Mix protocol if transferring eluates immediately after a purification
run.
In the Volume group, enter 5 in the Transfer field.
Select PCR Plate Type 1 from the Target Plate list. In the local tool bar the button is enabled.
Click the button. The Save Order dialog box opens and a default file name
<purification>_Transfer<n> is displayed indicating:
Click Save. The dialog box closes and the order file is saved in the database. The button is
enabled.
If you have modified an existing order and enabled the request for a change log in the ‘Settings’
dialog, you are prompted to describe the changes.
File Name Part Meaning
<purification> Name of the purification protocol the eluates were isolated with.
Transfer Indicates the volume transfer run.
<n> Number of the volume transfer run using the same plate derived
from the barcode.
