The message “PAUSE” is shown in the display and alternates with the elapsed time.
5.6.6 Continue program
To continue paused program press Soft Key [Continue]
5.7 Stop program
To stop the active program press [stop] Button.
To abort the active program confirm with [Yes].
5.7.1 Stop Program from a pause
If the last step in a program is a pause, the program has to be stopped by pressing the [Stop]
Button before switching off the instrument.
Note: If the instrument is switched off during a pause step, this will be recognized as a power
failure (a referring error message is written to the log file).
5.8 Optimization of Fast-PCR programs
The usage of Fast-PCR Kits allows a significant shortening of PCR protocols in comparison to
normal programs when conventional polymerases are used. Though suppliers of Fast-PCR kits
suggest some protocols for their polymerases for the optimization of Fast-PCR protocols some
physical factors have to be considered. E.g. the temperature transfer from the thermocycler
block into the reaction mix is hindered by low temperature conductivity of the plastic of the
reaction tubes. Moreover the temperature spread within the block, especially at short dwells,
depends on its geometry and is different between various block types. To reach the desired
temperature in the reaction tube, when dwells are shortened (
15 sec), the target temperature
of program steps has to be adapted, otherwise drop-outs due to unsufficient denaturation or
unsufficient primer annealing may occur. Biometra recommends the following protocol
templates for the optimization of dwells for the TProfessional Basic Thermocycler:
