Adjusting area scaling
Introduction This topic describes how to manually adjust the area scaling on
your cytometer if necessary for your application. The area scaling
is automatically set in CS&T. Depending on the size of your target
particle, you might need to adjust the area scaling manually. Larger
particles are more likely to require an area scaling adjustment.
About area scaling The area of a pulse is calculated by BD FACSDiva using measured
height and width measurements. It is sometimes important to
verify that the area calculation and the height measurement are
equivalent by adjusting the factor applied to the area. The required
area scaling factor changes based on sheath pressure and particle
size.
About this example The following example describes how to adjust area scaling for an
experiment which uses only the 488-nm laser and the 640-nm
laser. You must adjust area scaling for all lasers used in your
experiment. To adjust the other lasers, add a parameter and the
corresponding plots from that laser to the procedure.
Procedure To adjust area scaling:
1. Open an existing experiment, or create a new experiment in
the Browser.
2. Create a new specimen by clicking the New Specimen button
on the Browser toolbar.
