Purpose of This
Guide
This manual is written for principal investigators and laboratory staff responsible for
installing and maintaining the Applied Biosystems 7300, 7500, and 7500 Fast Real-Time
PCR Systems. This manual is designed to supplement the:
• Applied Biosystems 7300/7500/7500 Fast System Computer Setup Guide.
• Applied Biosystems Real-Time System Setup wizard, accessible from the
Installation CD or by selecting Program Files 7300/7500/7500 Fast
System Real-Time System Setup Wizard.
• Applied Biosystems 7300/7500/7500 Fast System Setup and Maintenance Video
CD provided with the instrument.
Audience This guide is intended for novice and experienced Applied Biosystems 7300, 7500, and
7500 Fast Real-Time PCR System users who need to install, maintain, or troubleshoot
their system.
Assumptions This manual assumes that you:
• Are familiar with the Microsoft
®
Windows
®
XP operating system.
• Understand general techniques for preparing and handling DNA samples.
• Have a general understanding of hard drives and data storage, file transfers, and
copying and pasting.
Text Conventions This guide uses the following conventions to make text easier to understand:
• Bold indicates user action. For example:
Type 0 and press Enter for the remaining fields.
• Italic text denotes new or important words and is also used for emphasis. For

Before performing a run, you must calibrate the instrument by performing the ROI,
background, and pure spectra calibrations.
• A right arrow () separates successive commands you select from a drop-down or
shortcut menu. For example:
