Problem Probable Cause Remedial Action
After switching from the
loading pump flow to the
nano flow, there is a drop in
the UV signal, and the signal
does not return to the initial
level fast enough.
There is dead volume in the
connection to the trap column.

The trap column is defective. Replace the trap column.
No peaks or only few, poorly
resolved peaks in the
chromatogram
The trap column is too short, the
flow rate of the loading pump is
too high and/or the loading time
is too long. (The three items
interact: If the trap column is too
short, peak may be lost because
they are not retained. If the
loading time is too long or if the
loading flow is too high,
hydrophilic peaks may be lost.
This also affects the
reproducibility because the
proteins are transported a long
way into the trap column.)
Consider the three items. Use a
longer trap column, reduce the
flow rate from the loading pump
and/or reduce the loading time.
Reproducible ghost peaks in
the chromatogram.
The degasser channels are
contaminated.
Rinse the degasser channels
( Solvent Rack manual).
The solvents are degraded or
dirty or their purity is
insufficient.
Use fresh solvents.
Contamination occurs
somewhere in the system.
Flush the system using an
appropriate solvent.
The temperature in the flow
manager does not change for
some time although the
temperature set point has not
been reached.
The ambient temperature is too
high.
Reduce the ambient temperature
(e.g., by ventilating the room).
The ventilation slots on the sides
and/or bottom of the instrument
are obstructed.
Make sure that the ventilation
slots are not obstructed in any
way.
The front panel door is not
completely closed.
When closing the door, make
sure that the locking mechanism
locks the door.
The capillaries are placed in such
a manner that ambient air can
enter the column chamber.
Make sure that the capillaries rest
flat on the edge of the housing.
The door seal is damaged. Contact Dionex Service.
