Setting up an experiment
4.1 Process selection
Select an appropriate cell isolation process depending on the desired target cell fractions, isolation strategy, and
column type. The last cell isolation process performed on the MultiMACS Cell24 Separator Plus is shown on the
upper left of the screen.
The MultiMACS Cell24 Separator Plus has five pre-defined processes.
l DEPLETE for the collection of unlabeled cells (negative fraction)
l POSSEL and POSSEL_SCA for the collection of labeled cells (positive fraction)
l POSSEL2 and POSSEL2_SCA for the collection of labeled and unlabeled cells (positive and negative fraction)
Use POSSEL and POSSEL_2 for separations with the Multi-24 Column Block and POSSEL_SCA and
POSSEL2_SCA for separations with MACS Columns in the MultiMACS Single-Column Adapter. DEPLETE can
be used with both, either Multi-24 Column Block or MACS Columns in the MultiMACS Single-Column Adapter.
The process selection chart in Figure 4.1 helps you to find a suitable program.
When using the processes POSSEL and POSSEL_SCA, the equilibration buffer and the unlabeled cells are
collected in the same container. Select DEPLETE, POSSEL2, or POSSEL2_SCA if you want to use the
unlabeled cells for downstream applications.
