4.4 Buffer volumes
Apply the following buffer volumes according to the column type and workflow.
Column type Equilibration Wash Elution
Multi-24 Column Block 1×2mL 3×1mL 1×1mL
LS Column 1×3mL 2×2mL 1×4mL
LD Column 1×2mL 2×1mL 1×3mL
Whole Blood Column 1×3mL 2×2mL 1×4mL

4.5 Large sample volumes
When exceeding a sample volume of 5mL, load the sample in two steps onto the column. Two 44.0mm deep
well plates are required for the collection of equilibration buffer, flow through, and wash. Alternatively, use a
88.0mm deep well plate.
1 Apply the first 5mL of the sample and let it run through.
2 Tap MOVE BACK.
3 Exchange the deep well plate after movement of the MultiMACS Cell24 Magnet.
4 Tap OK to proceed with sample loading.
Make sure that the column reservoir is empty before applying new buffer or samples.
4.6 Magnetic labeling
For details regarding magnetic labeling, follow the instructions in the MACS Cell Separation Reagent data sheet.
