Switch from Phase Contrast Microscopy to Epi-Fluorescence Microscopy
Epi-fluorescence and phase contrast methods can be used concurrently to cover the shortcomings of each. For
example, when locating the target, phase contrast microscopy can be used instead of epi-fluorescence
microscopy which tends to cause decoloration of the specimen.
This section provides tips for switching from phase contrast microscopy to epi-fluorescence microscopy when
devices required for epi-fluorescence microcopy and phase contrast microscopy are attached. Precautions on
concurrent use of the two methods are also noted. Also see “6 Phase Contrast Microscopy Procedure”.
(1) When turning on the power switch at the beginning of “6
Phase Contrast Microscopy Procedure”, keep the
power of the HG precentered fiber illuminator for
epi-fluorescence illumination off.
Check that the epi-fluorescence illumination path
shutter is closed.
The shutter is closed when the [FL SHUTTER] button is
lit.

(2) The position of the condenser turret is set to [OPEN]
bright-field position in procedure 6 in “6 Phase Contrast
Microscopy Procedure”, but

If the FL/DIC analyzer slider is in the optical path,
remove it from the optical path.
The slider is removed from the optical path by
pulling it out by one click-stop from the depressed
state.
Focus on the sample by bright-field and adjust the
optical system in this state.

epi-fluorescence microscopy.
