Optional: If using magnetic stirrer, insert the sterilized magnetic stirring bar into the vessel.
Optional: Install other sterilized sensors such as O
2
and CO
2
sensor if required.
8. Close the remaining open vessel ports with autoclaved screws.
9. Connect all tubing for gas inlet and gas/medium outlet and sampling as described in Fig. 14.
10. Close all openings, except the one for medium outlet, using sterile connectors.
11. Take the assembled vessel out of the hood and place it in the PBR, lock it with the side clips. Connect the
pH/temperature probe with the PBR (as described in chapter 4.3.1).
12. Note: It is recommended to let the medium in the vessel reach the desired cultivation temperature before
proceeding with the next step (calibration).
13. Prior to start of each experiment OD calibration should be performed with the medium used in that experiment.
Important Note: It is important to cover the front side of the vessel with the dark LED panel shield
(provided with the system) for the OD calibration to avoid possible interference of external light.
OD calibration can be performed manually on the front panel of the FMT150 control unit or by PBR software.
Please refer to chapter 7.1 on page 62 for manual control or to the PBR Software manual for calibration via
software.
14. After performing OD calibration add the prepared inoculum solution up to the desired OD value to start the
cultivation. Perform this step in the sterile environment of the flow hood.
Optional: User can employ a peristaltic pump to gradually introduce the inoculum into the vessel while monitoring
OD of the culture until the desired value is reached for optimal culture initiation.
15. Return the vessel with the culture to the photobioreactor.
16. Connect the vessel with the air line as described on page 32 (Fig. 19).
17. Note: If optional accessories are used follow the instructions for their installation.
18. Configure the experimental protocol in the PBR software and start the experiment. Detail description of these
steps are available in PBR Software manual.
Note: When using the photobioreactor without the PBR software, follow the manual instructions for
the setup of the growth parameters using the front panel of the PBR FMT150 control unit and as
described in chapter 7.1 on page 62.
