5.4 Prime to remove air from the pump and flow path
I
ntroduction
This section describes the recommended procedure on how to remove air from the
pump and flow path. The procedure is necessary to perform when a previously un-
used inlet is connected, and/or when changing the tank to a previously used inlet.
In UNICORN, use the PumpPrime instruction to remove air from the pump and flow
kit tubing with the pump prime function set to Run, e.g., PumpPrime →Run.
For other control software, refer to the software documentation to perform similar
procedure.
Note: If ÄKTA ready XL is equipped with two pumps and the gradient setup, make
s
ure to remove air from one line and pump at a time.
Prepare for priming and leakage
test
Follow the instruction below to prepare the system for priming.
Tip: If your process requires more inlets than the six available inlets, you can fit
a
Y- or T-tube to any inlet except for Inlet A1.
Step Action
1 Check that the inlet and outlet tubing are correctly connected.
2 Check that the buffer tanks are filled with a sufficient amount of a suitable
l
iquid for the procedure.
3 Check that valves to the buffer tanks are open.
4 Optional: When the tubing from the buffer tanks are connected to the cor-
responding inlets, carefully open each TC clamp prior to the system inlet
valve connection to allow for the air to escape and to fill the tubing as much
as possible. When the liquid has reached the inlet connection, tighten the
TC clamp again.
Note:
This procedure is performed to avoid pumping more air into the system. If
this is not applicable for the planned process, longer priming procedures to
remove air can be expected.
5 When applicable, check that the air sensor is connected at the inlet that
w
ill be used as the sample inlet.
