5. Box size increases resolution while maintaining the field of view. Increasing the zoom without
changing the box size will decrease the field of view.
6.13 Size of Pixels
The magnification and pixel size of an image is a function of lens magnification, box size and zoom
factor. However, these values will change slightly after any major alignment of the instrument.

The size of a pixel in a zoomed image is obtained by dividing the value from the above table by the
zoom factor. Shifting from a 512x512 to a 1024x1024 box size has the same effect on pixel size as
doubling the zoom factor. However, the 1024x1024 box will record a larger field of view, which may be
crucial for a given experiment.
Pixel Size = (Pixel size, Zoom = 1)/zoom factor.
e.g., 40X objective, 512x512,
zoom = 1, pixel size is 0.621 µm;
zoom = 4, pixel size is 0.621/4 = 0.155 µm/pixel
Size of image =(Box size) X microns per pixel.
e.g. 512 x 512 box, imaged with 20X objective, zoom = 1:
(512 pixels)(1.242 micron/pixel) = 635.904 microns width and height;
Divide this by the zoom factor to compensate for zoom.
