What to do if the fluorescence image is not visible on Olympus FLUOVIEW FV300?

If you cannot see a fluorescence image, start by verifying that the laser is oscillating by turning the laser unit ON and ensuring the emission key is set to ON. Confirm that the light path selector is correctly set to the LSM light path. Check for any obstructions in the light path, such as an engaged excitation tube or analyzer; if present, disengage them. Ensure the objective and confocal aperture knob are at their click positions. Verify that the confocal aperture is sufficiently open by increasing its size. Also, check the positions of the barrier filters and the light path detection switch, ensuring they are correctly set. Finally, verify that the fluorescent dye is compatible with the selected laser.
