This user’s manual is for the software to be run on Olympus FLUOVIEW FV300 Confocal Laser

Scanning Biological Microscope. To ensure safety, obtain optimum performance and familiarize yourself

fully with this product, we recommend that you study this manual thoroughly before operation.

This user’s manual is composed of two volumes including “SYSTEM OVERVIEW”, “OPERATION

INSTRUCTIONS”, “MAINTENANCE” and “TROUBLESHOOTING”. Together with this manual, please

also read the “User’s manual FLUOVIEW FV300” and the instruction manual of the microscope in order

to understand overall operation methods. To ensure the safety operation of laser system, we

recommend you to study the manual of each laser and the light source equipment besides this manual.

Retain this manual in an easily accessible place near a system for future reference.

  If you cannot see a fluorescence image, start by verifying that the laser is oscillating by turning the laser unit ON and ensuring the emission key is set to ON. Confirm that the light path selector is correctly set to the LSM light path. Check for any obstructions in the light path, such as an engaged excitation tube or analyzer; if present, disengage them. Ensure the objective and confocal aperture knob are at their click positions. Verify that the confocal aperture is sufficiently open by increasing its size. Also, check the positions of the barrier filters and the light path detection switch, ensuring they are correctly set. Finally, verify that the fluorescent dye is compatible with the selected laser.

Why can't I see a fluorescence image on my Olympus FLUOVIEW FV300 Microscope?

  There could be several reasons why you are unable to see a fluorescence image. First, ensure the laser unit is turned ON and the emission key is in the ON position, as the laser might not be oscillating. Also, check that the microscope's light path selector knob (or button on inverted microscopes) is set to the LSM light path. Another potential issue is an excitation tube or analyzer being engaged; disengage these from the light path. Confirm that the objective is correctly positioned at a click stop and that the confocal aperture knob is also at a click position, with the confocal aperture size appropriately increased.
