This user’s manual is for the software to be run on Olympus FLUOVIEW FV1000 Confocal Laser
Scanning Biological Microscope. To ensure safety, obtain optimum performance and familiarize
yourself fully with this product, we recommend that you study this manual thoroughly before operation.
This user’s manual is composed of two volumes including “OPERATION INSTRUCTIONS” and
“MAINTENANCE”. Together with this manual, please also read the “SAFETY GUIDE”, “HARDWARE
GUIDE” of User’s manual FLUOVIEW FV1000 and the instruction manual of the microscope in order to
understand overall operation methods. To ensure the safety operation of laser system, we recommend
you to study the manual of each laser and the light source equipment besides this manual.
Retain this manual in an easily accessible place near a system for future reference.

      If the laser is not output from the extremity of the objective, there could be several reasons. First, ensure that the laser unit is turned on and the emission key is set to ON. Next, verify that the correct laser wavelength is selected. For manual systems, check that the manual shutter of the fluorescence mirror unit is open and that the reflective mirror inside the fluorescence mirror unit is correctly positioned in the light path. Also, make sure the objective is in the light path, especially ensuring it's in the click position if using a manual revolving nosepiece. If the laser beam is too weak, increase the laser intensity. Lastly, confirm that the properties of the combined cube unit used for the ASU or OPD match the selected laser wavelength.

    Why can't I observe a fluorescence image with my Olympus Fluoview FV1000?

      If you cannot observe a fluorescence image, consider the following potential causes. The confocal pinhole diameter might be too small, so try increasing it. Ensure that the excitation Dichroic Mirror selection matches both the observed fluorescence wavelength and the excitation laser wavelength. Also, verify that the spectral dichroic mirror and barrier filter selections match the observed fluorescence wavelength. If you are using a spectral detection system, confirm that the acquisition wavelength region setting is suitable for the observed fluorescence wavelength. Check that the fluorescent dyeing method and excitation wavelength are compatible, selecting a laser optimum for the fluorescent dyeing method.
