Important: Please note that if the vessel is removed in 1000 ml or 3000 ml version of the PBR
FMT150, calibration constants stored in the system after OD calibration will no longer correspond to
the new conditions. In 1000 ml or 3000 ml PBR FMT150 version glass tunnel is used to improve
optical path for OD and fluorescence measurements, therefore any shift in the position in the vessel
might slightly change the optical path of the measuring beam and influence the absolute measured
values. Please refer to the OD calibration procedure for detail information.
4.2.2 MAINTENANCE
The whole vessel is fully autoclavable, including the detachable lid, connectors, sensors, and silicone tubing. However,
keep in mind that most materials deteriorate after repeated autoclaving. We recommend that you keep autoclaving to a
reasonable minimum. For most cases, hot water (60 – 70 °C), standard detergent and thorough mechanical cleaning are
enough to keep the equipment clean and non-contaminated. Optionally, the vessel and all the tubings can be disinfected
with 70% ethanol. When cleaning the cuvette, it is very important to rinse off any residue of detergent or medium and to
dry the cuvette with a soft cloth – this will help to prevent the cuvette corrosion. The cuvette should not come in contact
with strong acids or acid vapors.
The O
2
sensor and pH sensor may be autoclaved or sterilized up to 140 °C; the CO
2
sensor may be autoclaved or sterilized
up to 130 °C.
4.2.3 ASSEMBLY
Please refer to chapter 4.2.4 for details about description and assembly of 3000 ml vessel.
The next instructions describe cultivation vessel assembly:
• Remove the lid from the vessel by loosening the two screws with the provided hex key.
• Fill the vessel with medium, place the stir bar inside the vessel if used with the magnetic stirrer.
• Prepare the gas dispenser tubing by connecting the T-piece (n°16 in Tab. 4) with longer silicone tubing. Connect
the gas dispenser tubing to the two luer connectors as shown in Fig. 14A,B.
Standard version of the PBR FMT150 is connected according to the Fig. 13B, when optional modules
such as peristaltic pump or GAS and GMS are used see page 48 and 51 for further instructions).
• Close the last Luer Lock connector with the Luer Lock Ring Plug (n°4 in Tab. 4).
• Re-attach the lid to the vessel (Fig. 13C-3) using provided hex key. Make sure that the rubber gasket between the
vessel and the lid is properly installed and a good seal is achieved.
If needed, autoclave the assembled vessel.
