Nucleic Acids:Absorbance
The following information is required in order to save a new dye to the list:
Dye name
Analysis wavelength
Analysis wavelength specific molar absorption coefficient (M

)
260 nm absorbance correction factor
280 nm absorbance correction factor
Preconfigured dyes may not be modified or deleted.
Baseline Correction for Dyes
The software anchors the visual spectrum display to the baseline at 750 nm for
all measurements. It automatically applies a 340 nm bichromatic normalization
for the A260 value and uses a sloping baseline between 400 and 750 nm for dye
concentration calculations.
If adding a new dye, it is important to empirically determine if the default 750 nm
selection is appropriate to use as the baseline correction wavelength.
The baseline correction screen is accessed from the Run screen top action bar
Overflow icon.
Tap the Add icon to include a user-defined option between 750 and 840 nm.
The new option will be used for both the the visual display and the sloping dye
baseline.
Once a baseline correction nm is selected, it is applied to all subsequent
measurements and is not dye specific. When changing dyes, always confirm
that the proper baseline correction is applied.$
