Advanced Solvent Concentration
Note: The Solvent Concentration function in the Advanced tab of the Echo Liquid Handler software uses units of
acoustic impedance (MRayl). In cases where there are two simple solvent fluids, Water and Glycerol,fluid concentration
can be correlated to acoustic impedance.
The Advanced Solvent Concentration feature should only be used in cases where the concentration for a source plate is known. In
that case, accuracy can be improved by specifying the concentration prior to performing liquid transfers. The concentration
information entered is used with each source microplate until one of the following occurs:
l Use predefined checkbox is cleared
l New concentration information is entered
l Echo Liquid Handler software is restarted
The Echo Liquid Handler software uses the MRayl unit of measurement for the solvent concentration value. The solvent
concentration value can be calculated via a quadratic equation if the user knows the percent glycerol in the solution.
For example, for xxxx_AQ_GPx plate types with only glycerol and water, common conversions are:
Solvent Concentration Percentage (%Water/%Glycerol) Solvent Concentration Value (MRayl)
100% Water 1.48
90% Water / 10% Glycerol 1.60
50% Water / 50% Glycerol 2.10
The solvent concentration values shown in the table) above are calculated using a quadratic formula and the percent of glycerol in
the solution. The table below shows the equation and values required to calculate the solvent concentration value.
