In the Experiment Manager, double-click the sample to be analyzed to make the
sample the Active Sample. A red arrow at the left of the sample indicates this
sample is the Active Sample.
NOTE
Data analysis can be conducted when the sample acquisition is in process. In this
case, a green flashing arrow will show at the left of the sample currently acquiring
data. For example, in Figure 71, sample T-Lym is the Active Sample for data
analysis, while sample NKB is currently acquiring data.

Create Plots and Gates
Firstly, we need to look at the CD45 PerCP vs. SSC plot to locate the target cell
population, i.e. the lymphocytes, which have relatively large PerCP signal but small
SSC signal. In order to create the CD45 PerCP vs. SSC plot, click the dot plot icon
or the density plot icon from the data analysis toolbar. The sample name will
show at the top of the plot. The default parameters for a new dot plot or a new
density plot is always FSC Height (FSC-H) as the X-axis parameter and SSC Height
(SSC-H) as the Y-axis parameter. In this example, the X-axis needs to be changed
to be CD45 PerCP-H. To do so, right-click the FSC-H on the plot and select Height
> CD45 PerCP-H from the drop-down list. The X-axis parameter will change to
CD45 PerCP-H accordingly (Figure 72).
