3.2.2 Check of EX Wavelength Accuracy
Take the following procedure for the check.
(1) Click the (analytical method) button on the measurement toolbar and set wavelength scan for
measuring mode under the [General] tab on the Method window. Then select the [Instrument] tab
and set the conditions given below. After the setting, click OK button.
Scan mode : Excitation
Data mode : Luminescence
EM WL : 0
EX Start WL : 420
EX End WL : 450
Scan speed (nm/min) : 60
Delay : 0
EX Slit : 2.5
EM Slit : 20
PMT Voltage : 950
Response (s) : 0.5
Replicates : 1
(2) Click the (Pre Scan) button on the measurement toolbar. A pre-scan will thus be executed.
(3) Click the (start measurement) button on the measurement toolbar.
When a spectrum is acquired, carry out peak detection and determine the peak wavelength.
Make sure it is within the specification.
Specification: 435.8 1.5 nm
NOTE: If the photometric value obtained under the abovementioned conditions is excessive
(***.* is indicated for the photometric value) due to brightness of the room, try changing the
photomultiplier voltage to 700 V.
