3.5 Substage variant

The substage variant of the multiphoton detection system uses a high-NA oil-immersion condenser as a light
collector instead of a microscope objective. This condenser can also be used for illuminating the sample for
conventional imaging, and the system is provided with manual X, Y adjustments and a motorised focus
adjustment (on the SliceScope). The input dichroic mirror can be withdrawn from the optical path to permit the
use of visible light illumination if required (using the front-panel lever control).
Installation of the substage variant involves fitting the mounting bracket onto the SliceScope’s condenser focus
plate and then fitting the condenser to the optics block.
NOTE: Although the main optics block appears identical to the above-sample variant, there are internal
differences which make it impossible for the substage variant to be used above the sample.
3.5.1 Preparation
Check that you have all the required parts:
o Substage variant of the detection module, comprising
Optical block fitted with special top plate to take condenser U-AAC.
Fitted suspension arm with X, Y adjustment motion and mounting bracket
Two M5 X 20 hex cap-head bolts to fit the bracket to the SliceScope.
o Olympus U-AAC oil-immersion condenser (NA 1.4)
