2.4.3 Phase contrast
Prepare the microscope as described in Sections
2.4. and 2.4.1.
NOTE Phase-contrast objectives are labeled
in green.
Swing phase-contrast objective (2Ć15ā/ā3) in the
light path.
Open the aperture diaphragm (2Ć15ā/ā2) fully.
Insert the eyepiece telescope (2Ć15ā/ā4) in one of
the tubes in place of the eyepiece. Focus its eyeĆ
lens to make the phase rings of the objective visĆ
ible.
The phase ring of the phase-contrast objective apĆ
pears as a gray ring in the bright pupil (2Ć16ā/āA).

Slide the annular diaphragm (2Ć17ā/ā1) of a slider with phase diaphragm positions (2Ć15ā/ā1) matching the
objective and the inserted condenser in the light path.
The phase ring of the objective appears centrally in the field of view, while the bright annular stop may
be off-center (2Ć16ā/āB).
Turn the centering screws (2Ć17ā/ā3) on the slider until phase ring and annular diaphragm are superimposed.
The adjustment is correct, if the gray phase ring of the objective completely covers the bright ring stop
(2Ć16ā/āC).
