Other potential causes include the medium being packed too hard (repack the medium), blocked column nets (change nets), a blocked support net (clean or change the support net), an incorrect flow valve (change flow valve), or an incorrectly selected flow cell (select adequate size of flow cell). Also, check to see if the sample and collection vessels are at approximately the same level as, or above, the pump.

Also, check the column packing, equilibrate the column, and check the sample and sample handling to ensure old sample hasn't mixed with new.

The end-piece and/or flange might not be properly positioned with respect to the tube; in this case, disassemble and re-assemble the column. The solvents used might not be compatible with the O-rings and gaskets, so check solvent. The tubing might not be compatible with all the solvents, so change solvent. The connectors might not have the same dimensions, so change connectors. Air might be leaking into the column from the suction side of the pump, so check that the connections to and from the pump are correct and tightened. The torque might not be correct on the rods or the flange, so check that the correct torque specification has been used during column assembly.

  If your Cytiva Laboratory Equipment column is infected or contaminated with pyrogens, it could be due to several reasons: the column bed might show signs of bacterial growth, so you should perform CIP and prepare new buffers/sample. The starting material might be contaminated, so use clean material. The in going components (such as water, serum components in growth media, etc.) might not be ok, so check material. If positive LAL tests were seen after a CIP cycle with NaOH, it might be because the NaOH has been removed, so repeat cleaning.
