EDIT POLYMERASE
In the next screen (see Figure 40) touch the corresponding input boxes to enter a
name for the polymerase and set parameters for all requested program steps. With
two exceptions for most program steps the temperature and the holdtime must be
defined:
The temperature for the annealing step depends on the calculated primer melting
temperature (T
m
-value) and has to be set individually for each primer pair (see
chapter "Protocol Wizard parameter screen" p. 65).
The holdtime for the elongation step becomes calculated by the expected product
length (see chapter "Primer Annealing Temperature Calculator" p. 66).
The corresponding input boxes for the annealing temperature and elongation time are
shown in grey and cannot be activated.

After creating the program template for a new polymerase or editing the program
template of a polymerase press:
DONE
In the next screen (see Figure 41) touch the corresponding button to select a storage
place for the program template (polymerase). If the number of polymerases is higher
than can be maximal displayed in the list, the buttons above and below the polymer-
ase list can be used to scroll up and down.
