Values calculated from the regression line in standard curves, including the R

slope, and y-intercept. You can use the regression coecients to evaluate the quality
of results from the standards. See also “standard curve“ on page 191.
In standard curve and relative standard curve experiments, the best-t line from the
standard curve. Regression line formula:
C
t
= m [log (Qty)] + b
where m is the slope, b is the y-intercept, and Qty is the standard quantity.
See also “regression coecients“ on page 190.
An action that the software performs during analysis to remove one or more wells
from further analysis if a specic ag is applied to the well.
A method to determine relative quantities. This method compensates for target and
endogenous control eciency dierences within each run. In all experiments,
unknown samples and dilution series of template (such as cDNA) are amplied.
Following a run, the instrument software interpolates relative quantities for each
unknown sample from the appropriate dilution curve, then normalizes the data for
each sample (or set of replicates) as follows: target QAvg. ÷ endogenous control
QAvg.
A user-dened biological grouping. A replicate group may be a set of identical
reactions in a experiment.
Total number of identical reactions containing identical components and identical
volumes.
A uorescent dye used to detect amplication. With reagents, the reporter dye is
aached to the 5′ end.
An oligonucleotide that anks the 3′ end of the amplicon. The reverse primer and the
forward primer are used together in PCR reactions to amplify the target.
An enzyme that converts RNA to cDNA.
See “normalized reporter (Rn)“ on page 188.
A dye supplied by Thermo Fisher Scientic, which is used as a passive reference in
some experiments.
Denition of the reaction volume and the thermal prole for the instrument run. The
run method species the temperature, time, ramp, and data collection points for all
steps and stages of the instrument run.
The biological tissue or specimen that you are testing for a target gene.
