CyTOF XT User Guide 85
Clean the Autosampler Fluidic Lines and Nebulizer with
10% Bleach
Clean the Autosampler fluidic lines and nebulizer weekly with 10% bleach solution.
WARNING Health hazard. Turn down the nebulizer gas to 0.02 L/min to prevent
bleach aerosols from escaping the nebulizer rest and possibly being inhaled.
WARNING Health hazard. Do not mix bleach solution with an acidic solution
(such as Maxpar Washing Solution) because this mixture produces chlorine gas.
IMPORTANT Never run bleach solution when the nebulizer assembly is attached to the
heater assembly.
1 If plasma is on, turn it off.
2 Place the nebulizer in the nebulizer rest.
3 Turn down the nebulizer gas flow to 0.02 L/min.
NOTE At this flow rate liquids do not aerosolize, but it prevents liquid from building up
within the nebulizer. The liquid accumulates on the tip of the nebulizer and then drips
down.
a In CyTOF Software, on Instrument Configuration, click Ion Optics.
b Under Settings > Nebulizer Gas Flow, enter 0.02 and click Set.
4 Replace the 15 mL tube that is on the nebulizer rest with another 15 mL tube labeled
Bleach Waste.
5 Prepare a solution of 10% bleach by diluting 1 part household bleach with 9 parts DIW.
NOTE Prepare fresh 10% bleach solution each day that this procedure is performed.
6 Using 2 sample tubes, load at least 1 or 2 mL of 10% bleach solution in Location 1 and
DIW in Location 2 of the carousel.
7 Change the autosampler settings.
a In CyTOF Software, on Instrument Configuration, click Autosampler.
