3. Bring up the liquid level in "low" wells by adding HPLC-grade water with a pipette.
Determining the
aspirate offset
During a dispensing run, the pins dip into the microtiter plate wells to aspirate liquid. The
pins must dip far enough into the liquid to properly aspirate; however it is important that
the pins do not dip so far as to touch the resin in the wells. Optimally, the pins should dip
down to 0.5 mm above the level of resin.
The distance to which the dispensing pins are dipped is called the aspirate offset.
To determine the aspirate offset:
1. Tap the aspirate/dispense tab on the Method screen.

2. Leave the control software at this screen.
In the next two steps, you will measure the microtiter plate resin level. You will then
come back to this screen to enter an aspirate offset value.
3. Find the microplate well with the highest level of resin. Using a ruler or other
measuring device, measure how high (in mm) the resin goes from the bottom of the
well (Figure 2.30).
Most wells will have uniform levels of resin. There may not be an obvious well with
the highest level of resin. In this case, choose any well that is representative of the
highest level of resin in the wells.

Since this well has less resin, the liquid level
above it is lower than in the other wells, even
though the volume of liquid may be the same.
