Background
Fluorescence
Fluorescence data collected by the Applied Biosystems 7300/7500/7500 Fast Real-Time
PCR System includes a fluorescent signal inherent to the system, commonly referred to
as background fluorescence. Background fluorescence is a composite signal found in all
spectral data. This signal consists of fluorescence from several sources, including:
Background electronic signal
Contaminants in the sample block
The plastic consumable (plates and caps)
Guidelines for
Calibration
For a new instrument:
Always start with a new calibration kit.
Make sure the centrifuge you use is clean. Before centrifuging, wipe down the
bucket using a tissue.
Handle the calibration plates with care to prevent contamination. Do not place
plates on a lab bench, which may contaminate the plate. Always put calibration
plates back into their original bags.
