5.8.3 Fractionation overview
Fractionation types
The table below lists the types of fractionation that the fraction collectors can be used for.
DescriptionType
During fixed volume fractionation the fraction collector continuously
switches tubes according to the set volume throughout the entire fraction-
ation. This type of fractionation is also known as straight fractionation.
Fixed volume fractiona-
tion
Peak fractionation can be used to further increase the purity of the collect-
ed protein peaks and minimize the number of tubes used. The monitor
signal is used to determine when to switch the tubes. See Section 9.8.10
System settings - Watch parameters, on page 463 for information about
different watch options.
Peak fractionation
The two fractionation types listed above can be used in combination.
Combination of fixed volume and peak fractionation allows fractions col-
lected by fixed volume fractionation and fractions collected by peak frac-
tionation to be directed to different collection tubes.

To be able to analyze different parts of the peak, the fraction size during elution is usually set to a value
smaller than the expected peak volume.
