8.3.3 Preparations
1) Set the Chromatopac.
ATTEN: 10 WIDTH: 1 SPEED: 10
2) Turn the RF-20A(XS) ON with the [Func] key held down.
3) Set the photomultiplier applied voltage.
-HV SENS1: 600 -HV SENS2: 400 -HV SENS3: 200
4) Turn the pumps ON to start delivery of solution from all pumps. Flowrate 0.5 mL/min
5) Select the high-pressure valve to deliver 0.1 N sulfuric acid to the RF-20A(XS) side.
6) Start sensitivity compensation on the RF-20A(XS) after at least 30 minutes of pump
operation.
8.4 Adjusting SENS 1
1) Connect the RF-20A(XS) to the jig, and deliver 0.1 N sulfuric acid to the RF-20A(XS).
2) Turn the RF-20A(XS) ON with the [Func] key held down, and light the Xenon lamp.
(LAMP: 1)
3) Set up the RF-20A(XS).
MODE: 1 (1-wavelength measurement) EX: 350 nm EM: 450 nm
SENS: 1 (high) AIN: 1 (1) RESPONSE: 3 (0.5 sec)
CELL TEMP: 30 (RF-20AXS only)
RATIO: 1 (division) ANALOG1 MODE: 0 (CH1 integrator)
4) Start compensation at least one hour after lighting of the Xenon lamp on the RF-20A(XS).
5) Make sure that there are no air bubbles in the cell.
6) Start recording of the Chromatopac.
7) Execute RF-20A(XS) zero position adjustment. ([ZERO] key)
8) Calculate the sensitivity compensation value.
[RF-20A] Measure the room temperature (C) and calculate by the following formula:
Sensitivity compensation value = 600 { 1 - 0.005 (room temperature -
20) }
[RF-20AXS]
Sensitivity compensation value = 570
9) Make sure that 0.1 N sulfuric acid is being delivered and that the baseline has stabilized.
(Changes in fluorescent intensity value must be within 3/30 sec.)
10) Display "SENS." at "FACTORY", and press the [Enter] key.
11) When "SENS No." is displayed, enter "1" (high sensitivity), and press the [Enter] key.
12) When "SENS B ___" is displayed, make sure that the baseline has stabilized by judging
the displayed fluorescent intensity, and press the [Enter] key.
13) When "SENS INPUT" is displayed, enter the sensitivity compensation value calculated
earlier, and press the [Enter] key.
14) Select the high-pressure valve, and deliver 0.005 μg/mL quinine sulfate/0.1 N sulfuric
acid to the cell.
15) When "SENS K ___" is displayed, make sure that changes in the signal have stabilized,
and press the [Enter] key.
16) Automatic sensitivity compensation is executed ("SENS.../Adjusting" is displayed), and
after execution ends, the photomultiplier applied voltage of SENS 1 "SENS HV ___" is
displayed. Note down the photomultiplier applied voltage, and press the [CE] key.
17) Select the high-pressure valve, and deliver 0.1 N sulfuric acid to the cell. (for three
minutes or more)
18) Repeat steps (7) to (16).
8.4.1 Verifying the Sensitivity of SENS 1
1) Set up the RF-20A(XS).

2) Start recording of the Chromatopac.
3) Make sure that 0.1 N sulfuric acid is being delivered and that the baseline has stabilized.
(Changes in fluorescent intensity value must be within 3/30 sec.)
