4) Execute RF-20A(XS) zero position adjustment. ([ZERO] key)
5) Select the high-pressure valve, and deliver 0.005 μg/mL quinine sulfate/0.1 N sulfuric
acid to the cell.
6) After making sure that changes in the signal have stabilized, note down the fluorescent
intensity value.
[Judgment] The fluorescent intensity value must be within ±5% of the sensitivity
compensation value.
7) Select the high-pressure valve, and deliver 0.1 N sulfuric acid to the cell. (for three
minutes or more)
8.4.2 Adjusting SENS 2
1) Deliver 0.1 N sulfuric acid to the cell.
2) Set up the RF-20A(XS).
MODE: 1 (1-wavelength measurement) EX: 350 nm EM: 450 nm
SENS: 2 (medium) GAIN: 1 (1) RESPONSE: 3 (0.5 sec)
CELL TEMP: 30 (RF-20AXS only)
3) RATIO: 1 (division) ANALOG1 MODE: 0 (CH1 integrator)
4) Make sure that there are no air bubbles in the cell.
5) Start recording of the Chromatopac. (ATTEN: 9)
6) Execute RF-20A(XS) zero position adjustment. ([ZERO] key)
7) Calculate the sensitivity compensation value.
[RF-20A] Measure the room temperature (C) and calculate by the following formula:
Sensitivity compensation value = 375 { 1 - 0.005 (room temperature -
20) }
[RF-20AXS]
Sensitivity compensation value = 356
8) Make sure that 0.1 N sulfuric acid is being delivered and that the baseline has stabilized.
(Changes in fluorescent intensity value must be within 3/30 sec.)
9) Display "SENS." at "FACTORY", and press the [Enter] key.
10) When "SENS No." is displayed, enter "2" (medium sensitivity), and press the [Enter] key.
11) When "SENS B ___" is displayed, make sure that the baseline has stabilized by judging
the displayed fluorescent intensity, and press the [Enter] key.
12) When "SENS INPUT" is displayed, enter the sensitivity compensation value calculated
earlier, and press the [Enter] key.
13) Select the high-pressure valve, and deliver 0.1 μg/mL quinine sulfate/0.1 N sulfuric acid
to the cell.
14) When "SENS K ___" is displayed, make sure that changes in the signal have stabilized,
and press the [Enter] key.
15) Automatic sensitivity compensation is executed ("SENS.../Adjusting" is displayed), and
after execution ends, the photomultiplier applied voltage of SENS 2 "SENS HV ___" is
displayed. Note down the photomultiplier applied voltage, and press the [CE] key.
16) Select the high-pressure valve, and deliver 0.1 N sulfuric acid to the cell. (for three
minutes or more)
8.4.3 Verifying the Sensitivity of SENS 2
1) Set up the RF-20A(XS).
MODE: 1 (1-wavelength measurement) EX: 350 nm EM: 450 nm
SENS: 2 (medium) GAIN: 1 (1) RESPONSE: 3 (0.5 sec)
CELL TEMP: 30 (RF-20AXS only)
