7.4.2 Prediluted mode
The sample pre-dilute method test protocol has
been specifically developed to allow Elite 5 users
to measure CBC results on samples having only
small volumes or when one of the counted
parameter result is higher than its linearity range.
The sample must be diluted off-line in an
appropriate tube with physiological saline at a ratio
of 1 part blood sample to 5 parts saline. It is
important to ensure there is a sufficient final
volume in the tube to enable the analyzer to
aspirate the diluted sample satisfactorily. The
suggested minimum diluted sample volume is
300ul from Vacutainer.
When the pre-dilute mode is selected on the
analyzer all results are automatically corrected and
calculated for the dilution factor.
How to use the pre-diluted sample mode
Step 1. Prepare a diluted patient sample at a dilution ratio of 1:6 (1 part blood to 5
parts saline) in physiological saline. Please ensure that the sample is well mixed and
placed on the analyzer immediately after preparation.
Step 2. Select the pre-dilute mode on the analyzer
Step 3. Place the diluted sample onto the analyzer using the
appropriate tube holder
Step 4. Initiate the measurement by pressing start button.
Step 5. Analysis will take place. Please note that due to the lower cell numbers it is
necessary for analyzer to count for a longer period than in the normal sample mode.
(Approximate read time is 16 seconds as opposed to 8 seconds)
Step 6. CBC results are corrected for the dilution factor, displayed on screen and can
also be printed. Please note it is indicated on the screen and on the print-outs that the
measurement was conducted in the pre-diluted sample mode.
Step 7. Remove diluted sample and to move next measurement ensuring correct
sample type is selected.
Important notes
It is also important to use physiological saline and not analyzer diluent to prepare
diluted samples.
The pre-diluted sample option can only be used manually and not with the autoloader.
