7.7.1. Preparing blood sample for RBC aggregation test
A blood sample can be stored at a maximum of one hour (at room temperature). After that the sample
cannot be used anymore.
Since aggregation is determined through light back-scatter measurement and indicated as Isc-
values are dependent on the amount of oxygenated Hb present, it is important to use fully
oxygenated whole blood. When the degree of oxygenation is different in two samples of the same
blood, all absolute Isc-values, derived from the Syllectogram, will be different!
1. Incubate 4 ml of whole blood, with at least 40 ml of air (1:10), in a tube preferably on a roller-
bank for 15 minutes, minimum. About 600-900 µl of whole blood is required to perform an
aggregation measurement.
