Thermal Denaturation Mode 3
• The Ramping Speed group allows you to select the speed at which the
temperature is increased. Choose Slow to increase the temperature
incrementally to each step of the temperature ramp. Choose Fast to
increase the temperature ballistically to each step of the temperature ramp.
• Select Autosave spectra to file to specify that the acquired spectra are to be
saved. Type a name for the data file in the Autosave filename: field. The
name must comply with DOS naming conventions (maximum 8
alphanumeric characters). When Autosave spectra to file is cleared, the
spectra are not saved.
• You can type a description of the method in the Method Information field.
The field accepts any alphanumeric characters, and the description can be
as long as you wish. The text wraps automatically at the end of the line.
The Set Individual Calculation Range dialog box is displayed when you choose
the Set Individual Calculation Range command from the Method menu. It
enables you to define a temperature range for the calculation of thermal
denaturation results on the currently-displayed trace. You type the lower and
upper limits of the temperature range in the from and to fields in the dialog
box.
If the steps are large, there is a danger of the temperature overshooting in the Fast mode.
