Setting Up for Sorting
In general, do the following to set up for a sorting experiment. Each step is
explained in more detail in previous or subsequent sections.
1 Start up the cytometer, the computer, and perform cytometer QC.
• For startup, see Cytometer Startup on page 108.
• For cytometer QC, see Cytometer Quality Control Using BD
FACSDiva Software on page 118.
• If you are using the CS&T module for performance checking, see
Checking Performance with Cytometer Setup and Tracking on
page 117.
2 Install the appropriate size nozzle.
As a general guideline, the nozzle opening should be at least three times the
diameter of the particle to be sorted. See Changing the Nozzle on page 205.
3 Select a new sort setup option, if needed, or select an appropriate cytometer
configuration, including the sort setup mode.
To change your current sort setup mode, select a different setting from the
Sort Setup menu.

4 Check the laser delay for your sheath pressure and particle size.
• To set the laser delay automatically using the Cytometer Setup and

5 Optimize cytometer settings for the sample to be sorted.
