Setting Up for Sorting
In general, do the following to set up for a sorting experiment. Each step is
explained in more detail in previous or subsequent sections.
1 Start up the cytometer, the computer, and perform cytometer QC.
For startup, see Cytometer Startup on page 108.
For cytometer QC, see Cytometer Quality Control Using BD
FACSDiva Software on page 118.
If you are using the CS&T module for performance checking, see
Checking Performance with Cytometer Setup and Tracking on
page 117.
2 Install the appropriate size nozzle.
As a general guideline, the nozzle opening should be at least three times the
diameter of the particle to be sorted. See Changing the Nozzle on page 205.
3 Select a new sort setup option, if needed, or select an appropriate cytometer
configuration, including the sort setup mode.
To change your current sort setup mode, select a different setting from the
Sort Setup menu.

4 Check the laser delay for your sheath pressure and particle size.
To set the laser delay automatically using the Cytometer Setup and
tracking module, see Checking Performance with Cytometer Setup and
Tracking on page 117.
To set the laser delay manually, see Cytometer Quality Control Using
BD FACSDiva Software on page 118.
5 Optimize cytometer settings for the sample to be sorted.
