4-12 Rough-cut (roughing) mode
Roughing is a process performed to expose the surface of a tissue sample embedded in a block of
paraffin. In this process, slices are cut thicker than in the normal slide preparation process. The AS-
410M can perform roughing either as a single independent operation or as an intermediate part of the
slide preparation process.
This Chapter explains the operational steps for performing roughing as a single independent
operation. Unlike a roughing operation performed as a part of the slide preparation process, a single
roughing operation allows the User to check the exposed conditions of tissue samples block by block.
Follow the steps below to perform a roughing using the AS-410M:
1. Use the sample transfer robot to transfer a sample block from a sample tray to the sample holder.
2. Use the touch sensors to measure the height and angle of the sample surface. Then, use the
“tilting mechanism” to adjust the height and angle relative to the microtome blade.
3. Perform a predetermined number of times of roughing to cut slices of a predetermined thickness.
4. Roughing can be performed while visually observing the tissue surface exposed from the surface
of the sample block. Alternatively, the User can perform roughing and check exposure of the
tissue surface while attaching thin sliced sections to the carrier tape.
5. After the completion of the roughing, replace the sample block back onto the sample tray.
The auto surface detection (auto trimming) function is NOT available in the
roughing mode. Select the slide preparation mode to use the auto surface
detection function.
